Background

Decidualization is essential for embryo implantation.Obese-women have lowerpregnancy rates and higher rates of pregnancy loss than normal-weight women.However,Theeffect of obesity on decidualization is unclear.

Methods

Female C57Bl/6 mice were fed either a high-fat or a diet of standard chow for 12weeks.The number of implantation sites were analyzed in mice mated with normal males.In micemated with vasectomized males,decidualization was induced and both sides weight of the uteruswere assessed.Mouse ESCs were induced to decidualize in vivo by treatment with Estradiol andprogesterone in the presence or absence of oleic acid and palmitic acid or etomoxir.Lipidmetabolomics and proteomics was detected by mass spectrometry.The uterine tissues or cells wereanalyzed via qPCR,TEM and Western Blots.

Results

HF mice became obese.Compared with control mice,HF mice had smaller implantationsites and higher rate of infertility.In the artiļ¬cial decidualization experiments,mice exposed to theHF diet developed smaller deciduomas than CON mice.In HF group,protein expression ofdecidual markers was decreased in vivo and vitro experiments,the composition ofMedium-long-chain fatty acid was transformed.There were 105 proteins regulated in obesemice.Mitochondrial structure shows abnormal and increased lipid droplet in uterine tissues ofobese mice.Protein expression of CPT1a and FAS was increased,Protein expression of ACC andP-ACC was reduced in vivo and vitro experiments.Addition inhibitors of CPT1a can amelioratethe decidual impairment in vivo experiments.

Conclusions

Excess fatty acids that can accumulate as potentially toxic lipid intermediates arelinked to the impairment of decidualization by causing metabolic disorder of animals and cells.
